This review describes methods for the Agrobacterium-mediated T-DNA transfer to plant cells under in planta conditions. The main attention is focused on the techniques for gene transfer with Agrobacterium T-DNA to plant generative cells. The  effects of plant genotype, bacterial strain, type of vector constructs, composition of inoculation medium and conditions for plant treatment with agrobacteria on the efficiency of Agrobacterium-mediated transformation in planta have been analyzed. On the basis of the reported and authors’ own  experimental data, a putative mechanism of the plant generative cells Agrobacterium-mediated transformation in planta is discussed.
