The influence of heat-shock conditions (temperature and duration of exposure) and size of buds on the emryogenesis in a rape microspores culture has been investigated. It was shown, that size of buds has no effect on the formation of haploid plants because embryogenesis is a function of stage of pollen development which is in turn determined by growth condition for the original material. The optimal heat-shock protocol proved to be as follows: for Topaz cultivar, 35°С and 72 h, and for Centr 1 line, 35°С and 24 h. We have obtained 145 embryos, 50 flowering plants of Topaz cultivar, 36 of which turned to be haploid (haploidy frequency of 0,72) and 53 embryos, 16 flowering plants and 4 haploid plants of Centr 1 line (haploidy frequency of 0,25). The haploid nature of plants was proved by the cytological analyses.
Key words: Brassica napus, double haploids, haploids, microspore culture, oil rape.
