The efficiency of the secretion of the humanized light kappa-chain of the murine H3-1 antibody by the Pichia pastoris yeast as depending on the copy number of pro-regions in the leader pre-pro-polypeptides has been studied. The leader pre-pro-polypeptides were composed of the signal peptide (pre-region) from the Saccharomyces cerevisiae α-factor precursor and a variable number of pro-regions from the pre-pro-precursors of S. cerevisiae α-factor of Hsp150p, or P. pastoris Hsp150p. The increase in the pro-region copy number was shown to be accompanied by either near 1,5-fold growth or near 5,0-fold decrease in the secretion level depending on the pro-region used. It was concluded that enhancing of the pro-region copy number is of potential value in the intensification of the protein secretion in P. pastoris.
