A method for the in vitro assessment of the toxic effect of some human diseases causes as a result of their genes expression in human cells has been suggested. The technique is based on the transient expression of the genes of interest in the HEK293 line cells. Using this approach, it was demonstrated for the first time that the expression of human hepatitis A virus 3C protease induces cell death. It was also shown that the expression of human poliovirus 2A protease results in cell death, whereas cathepsins B and D did not show any cytotoxic effect on the used culture.

Key words:  cell death,  cytotoxicity,  lipofection,  transient expression.
