A new extracellular cyclodextrin glucanotransferase (CGTase) ( EC 2.4.1.19) has been found out in a bacterial species of Paenibacillus ehimensis and characterized. The enzyme from the P. ehimensis VKM B-2680D (IB-739) isolate was partially purified from culture liquid by the adsorption on starch and column chromatography on DEAE-Sepharose. The CGTase was stable within the pH range 5,5—8,2 and temperature inferior to 60C. The enzyme showed the maximal cyclizing activity at 40—50°C and pH 5,5—6,5. According to the SDS-electrophoresis data, the enzyme has a molecular mass of ~ 75 kDa. The CGTase thermal stability could be slightly improved in the presence of 10 мМ CaCl2. The daily conversion of 5% potato starch to α-, β-, and γ-cyclodextrins under the optimum conditions was 40% with their mass ratio equal to 14,6:21,4:4,0, respectively.
