Culturing conditions for a recombinant interferon-beta-1а producing cell line based on Chinese hamster ovary cells (CHO) and cumate expression system have been optimized. The addition of 10 mM galactose and 2 mM uridine to a media was shown to improve the glycosylation profile of the target protein. Scaling-up of the cultivation process to single-use wave-induced motion bioreactor allowed to maintain the cell growth rate, specific productivity and interferon-beta-1а glycosylation that are characteristic of the small-volume culturing.

Key words:  bioreactor,  CHO cells,  cultivation,  interferon-beta-1a,  glycosylation,  optimization.
