Metabolic engineering approaches including those used by the authors applied to the obtaining of Escherichia coli phenylalanine producers have been systematized in this review. For significant phenylalanine production, optimal strengthening of the phenylalanine biosynthesis from erythrose-4-phosphate and phosphoenolpyruvate and rationalization of the synthesis of these precursors were performed. In order to more completely re-orientate the metabolism to the phenylalanine production, a modification of global regulator mechanisms, in particular, the Csr (carbon storage regulator) system was investigated. The identification of an aromatic amino acid exporter YddG was made with the help of the phenylalanine producer as a test system. Original approaches to designing of a phenylalanine producer (the use of a leaky allele tyrA—ssrA, and phosphate regulon promoters), as well as new methods for the construction of producers of the related amino acid of tyrosine are discussed. The examples of the production of valuable aromatic compounds synthesized from phenylalanine or its precursor phenylpyruvate in E. coli phenylalanine producers as hosts for heterologous gene expression are considered.
